In order to measure the concentration of dsRNA, we should first measure standard 2Xpepper RNA sample and construct a standard curve between pepper RNA concentration and light strength of pepper RNA. 1.We first use DEPC water dissolve standard pepper RNA sample and adjust its concentration into a series of pepper RNA standard solution:0, 10, 20, 50, 100, 200, 500, 1000nM. 2.Then dissolved HBC and diluted it with HEPES buffer into 4μM. 3.Mix standard pepper RNA solution with 4μM HBC solution in a same volume and setting for 20min. 4.To measure the light strength of pepper RNA, we use spectrometer measure the mix fluorescent strength at specific wave length. 5.Setting 0 nM pepper RNA solution light strength as zero, we recorded pepper RNA fluorescent strength and plotted.

In our prospect, the fluorescent light strength will be measured. Here is the light strength that was described on the reference.
